Determination of dietary fiber content in bean curd residue with a fiber tester

In recent years, as people's living standards have increased, the intake of dietary fiber in food has decreased, causing the incidence of certain diseases of the digestive tract, cardiovascular diseases, hypertension, and obesity to increase year by year. This has caused widespread medical problems. Workers and food nutritionists attach great importance to it, and as a result, the development and utilization of dietary fiber has risen quietly. However, there are few reports on the methods for determining the dietary fiber that provide basic theoretical basis for the development and utilization of dietary fiber, either the operation is too cumbersome and takes a long time, or the measurement result is far from the theoretical value. Recently, we have done a research on the determination method of dietary fiber in bean curd residue. Now we report the relevant methods as follows. The fiber content was measured using a fiber meter.

Take 3 parts of gelatinized 0.5% soluble starch solution, each 50ml, add the amount of human amylase solution, a-amylase solution, and peak amylase solution respectively. Under appropriate conditions, incubate for a certain period of time until iodine is added. The liquid check is not blue. After cooling, each part was taken out, diluted appropriately, and the amount of reducing sugar was determined and converted into starch. The results showed that the amount of hydrolyzed starch was 56.43%, 58.78% and 64.97%, respectively. In the remaining starch hydrolyzate, add 45% of its 95% ethanol and leave it overnight at room temperature. The results of the fiber tester showed that the peak amylase was still clear and there was no precipitate; the others had obvious flocs suspended or sinking to the bottom of the bottle. It can be seen that the peak amylase has a strong ability to hydrolyze starch, the degree of hydrolysis is deeper, and production is not easy to be solidified by ethanol. pJ dextrin.

The content of dietary fiber in the bean dregs was found to be 63.00%, which was close to the theoretical value (63.92). The precision was high and the degree of dispersion was small. This shows that the method is well-designed and the measurement results are reliable. If the filtration is changed to centrifugal separation of sediment, not only the human error will be reduced, but also the operation will be easier and the measurement result will be more satisfactory.
